Answer the following questions based on the procedure of hydrolysis of carbohydrates. Write the observation and reaction of each solution. Explain your answers
Hydrolysis of Carbohydrates
1. Get three clean and dry test tubes. In the first test tube, mix 1mL of 3M HCl and 5mL 1% sucrose solution. Stir and heat the solution in a water bath for 20 minutes. Cool and add 1mL of 3M NaOH dropwise to make the solution neutral.
2. Transfer 10 drops of the solution to another test tube and add 2mL of Fehling’s A and B solution. Heat for few minutes. Record your observation.
3. In the second test tube, place 5mL of 1% starch solution and in the third test tube, place 5mL of 1% glycogen solution (from isolation of glycogen, experiment 4).
4. Add 5 drops of conc. HCl to each of the test tubes. Cover each tube with a marble. Boil the samples in a water bath for 30 minutes.
5. The boiled samples will be called the HYDROLYSATE SAMPLES of starch and glycogen used for the qualitative Benedict’s and Barfoed’s test.
Write the observation and reaction of each solution. Indicate the color and formation of precipitate in each solution. Explain your answer.
1. 1% SUCROSE
2. 1% STARCH
3. 1% GLYCOGEN
