A variety of methods have been used to measure plasma triglycerides. There are the stepwise colorimetric methods based on the Van Handel-Zilversmit reaction and Hantzsch reaction. The CDC reference method used a chloroform extraction procedure followed by silicic aci chromatography to isolate the triglycerides, then apply the Zilversmit reaction to measure the triglycerides concentration. But the methos most commonly used for clinical or epidemiologic purposes are based on the hydrolysis of triglycerides and the measurement of glycerol that is released in the reaction.
